¥°. Preparation of Antihuman Spleen Tissue Rabbit Serum


Abstract
Rabbits were immunized against human fresh spleen tissue with Frond¢¥s complete adjuvant. All of rabbits were immunized once for two weeks period.
The antibody titer of antisera were measured by leucoagglutinin method. The titers of all antisera were summarized in table 1. In order to get active antibody component in the serum, the fractionation of the antiserum was carried out with ammonium sulfate precipitation.
And it was confirmed by leucoagglutination that these precipitated fractions of antisera have the antibody activity.
And also these precipitated fractions were confirmed as almost entirely of gamma globulin by cellulose acetate electrophoresis (Fig. 1). When the antibody reactivity of these precipitated fraction was measured by leucoagglutinin test, these showed almost similar leucoagglutinin titer compared with leucoagglutinin titer of whole antiserum even a few of precipitate showed some what lower titer.
